The p.A1843T variant (also known as c.5527G>A), located in coding exon 10 of the BRCA2 gene, results from a G to A substitution at nucleotide position 5527. The alanine at codon 1843 is replaced by threonine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -

In summary, this is a novel missense change that is not predicted to affect protein function or cause disease. However the evidence is insufficient at this time to prove that conclusively. It has been classified as a Variant of Uncertain Significance. The threonine amino acid residue is found in multiple mammalian species, suggesting that this missense change does not adversely affect protein function. In addition, algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies. This variant has not been published in the literature and is not present in population databases. This sequence change replaces alanine with threonine at codon 1843 of the BRCA2 protein (p.Ala1843Thr). The alanine residue is weakly conserved and there is a small physicochemical difference between alanine and threonine. -
