The p.L3035I variant (also known as c.9103C>A), located in coding exon 62 of the ATM gene, results from a C to A substitution at nucleotide position 9103. The leucine at codon 3035 is replaced by isoleucine, an amino acid with highly similar properties. This variant was observed in 1/287 patients with hereditary breast and/or ovarian cancer; this patient was diagnosed with breast cancer at age 39 and had a family history of breast cancer (Caminsky NG et al. Hum Mutat, 2016 Jul;37:640-52).This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
