The c.4790C>A (p.S1597Y) alteration is located in exon 11 (coding exon 10) of the BRCA2 gene. This alteration results from a C to A substitution at nucleotide position 4790, causing the serine (S) at amino acid position 1597 to be replaced by a tyrosine (Y). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
Missense variant in a coldspot region where missense variants are very unlikely to be pathogenic (PMID:31911673). -
This missense variant replaces serine with tyrosine at codon 1597 of the BRCA2 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in individuals affected with breast or ovarian cancer (PMID: 22366370) and colorectal cancer (PMID: 28591715). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD) but has been reported in control individuals (PMID: 18844490). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

This missense change has been observed in individual(s) with a personal or family history of breast/ovarian cancer as well as healthy individuals (PMID: 18844490, 22366370).
