This variant has been reported in the published literature in individuals with personal or family history of breast cancer and/or ovarian cancer (PMIDs: 25863477 (2015), 32341426 (2020), and 33471991 (2021), see also LOVD (http://databases.lovd.nl/shared/genes/BRCA2)). This variant has not been reported in large, multi-ethnic general populations (Genome Aggregation Database, http://gnomad.broadinstitute.org). Based on the available information, this variant is classified as pathogenic.

The c.8639_8640delCA pathogenic mutation, located in coding exon 20 of the BRCA2 gene, results from a deletion of two nucleotides at nucleotide positions 8639 to 8640, causing a translational frameshift with a predicted alternate stop codon (p.T2880Nfs*26). This alteration was identified in a study of 2403 Korean breast cancer patients at risk for hereditary breast and ovarian cancer (Kang E et al. Breast Cancer Res. Treat., 2015 May;151:157-68). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
This variant deletes 2 nucleotides in exon 21 of the BRCA2 gene, creating a frameshift and premature translation stop signal.
