Variant summary
Our verdict is Pathogenic. The variant received 18 ACMG points: 18P and 0B. PVS1PM2PP5_Very_Strong
The NM_007294.4(BRCA1):c.3143delG(p.Gly1048ValfsTer14) variant causes a frameshift change involving the alteration of a non-conserved nucleotide. The variant was absent in control chromosomes in GnomAD project. Variant has been reported in ClinVar as Pathogenic (★★★). Synonymous variant affecting the same amino acid position (i.e. G1048G) has been classified as Likely benign. Variant results in nonsense mediated mRNA decay.

The BRCA1 c.3143del (p.Gly1048Valfs*14) variant alters the translational reading frame of the BRCA1 mRNA and causes the premature termination of BRCA1 protein synthesis. This variant has been reported in the published literature in individuals with breast cancer (BC) (PMIDs: 22762150 (2012), 32341426 (2020)) and in individuals with ovarian cancer (OC) (PMID: 36367610 (2023)). Based on the available information, this variant is classified as pathogenic. -
Hereditary cancer-predisposing syndrome Pathogenic:1
The c.3143delG pathogenic mutation, located in coding exon 9 of the BRCA1 gene, results from a deletion of one nucleotide at nucleotide position 3143, causing a translational frameshift with a predicted alternate stop codon (p.G1048Vfs*14). This pathogenic mutation was detected in one family undergoing BRCA1 and BRCA2 testing (Lecarpentier J et al. Breast Cancer Res. 2012;14:R99). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
