This premature translational stop signal has been observed in individual(s) with clinical features of hereditary motor and sensory neuropathy (PMID: 21507954, 30460542, 31692161). ClinVar contains an entry for this variant (Variation ID: 280809). This variant disrupts a region of the SACS protein in which other variant(s) (p.Asn4549Asp) have been determined to be pathogenic (PMID: 15156359, 21507954). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. We interpret L3102X as a pathogenic variant. -
