The observed missense c.604C>A p.Pro202Thr variant in MTHFR gene has been previously reported in individuals affected with MTHFR-related disorders Schiff et al., 2011; Froese et al., 2016. This variant is absent in gnomAD Exomes. This variant has been submitted to the ClinVar database as Pathogenic. Computational evidence Polyphen - Benign, SIFT - Damaging and MutationTaster - Disease causing predicts conflicting evidence on protein structure and function for this variant. The reference amino acid of p.Pro202Thr in MTHFR is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. The amino acid Pro at position 202 is changed to a Thr changing protein sequence and it might alter its composition and physico-chemical properties. However, additional functional studies will be required to prove the pathogenicity of this variant. For these reasons, this variant has been classified as Likely Pathogenic. -
