Variant summary: POLR3GL c.2T>C (p.Met1Thr) alters the initiation codon and is predicted to result either in absence of the protein or truncation of the encoded protein due to translation initiation at a downstream codon. Two of three in-silico tools predict a damaging effect of the variant on protein function. The next in-frame ATG (Met) is located in codon 71 (exon 3). No pathogenic variants upstream of the closest potential new in-frame start site have been reported in HGMD or ClinVar. In addition, loss-of-function has not clearly been established as the disease mechanism for this gene. The variant was absent in 158096 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.2T>C in individuals affected with Short Stature, Oligodontia, Dysmorphic Facies, And Motor Delay and no experimental evidence demonstrating its impact on protein function have been reported. No submitters have cited clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance. -
