Genes affected
CTSK (HGNC:2536): (cathepsin K) The protein encoded by this gene is a lysosomal cysteine proteinase involved in bone remodeling and resorption. This protein, which is a member of the peptidase C1 protein family, is predominantly expressed in osteoclasts. However, the encoded protein is also expressed in a significant fraction of human breast cancers, where it could contribute to tumor invasiveness. Mutations in this gene are the cause of pycnodysostosis, an autosomal recessive disease characterized by osteosclerosis and short stature. [provided by RefSeq, Apr 2013]

Pathogenic, criteria provided, single submitterclinical testingInvitaeOct 25, 2019This sequence change results in a premature translational stop signal in the CTSK gene (p.Arg312*). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 18 amino acids of the CTSK protein. This variant is present in population databases (rs375958814, ExAC 0.004%). This variant has been observed in the homozygous state or in combination with another CTSK variant in individual(s) with pycnodysostosis (PMID: 24767306, Invitae). In at least one individual the data is consistent with the variant being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 371426). For these reasons, this variant has been classified as Pathogenic. -
