Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 251470 control chromosomes (gnomAD). c.934C>G has been reported in the literature in individuals affected with Pyknodysostosis (e.g. Hou_1999, Fratzl-Zelman_2004). These data indicate that the variant is likely to be associated with disease. One of these reports also performed in vitro expression studies with the variant and found no detectable mature protein or enzyme activity ( Hou_1999). The following publications have been ascertained in the context of this evaluation (PMID: 10074491, 15070910). ClinVar contains an entry for this variant (Variation ID: 370960).
