Variant summary
Our verdict is Uncertain significance. Variant got 2 ACMG points: 2P and 0B. PM2
The NM_006118.4(HAX1):āc.14A>Gā(p.Asp5Gly) variant causes a missense change. The variant allele was found at a frequency of 0.0000062 in 1,613,880 control chromosomes in the GnomAD database, with no homozygous occurrence. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar. Another variant affecting the same amino acid position, but resulting in a different missense (i.e. D5A) has been classified as Uncertain significance.

The p.D5G variant (also known as c.14A>G), located in coding exon 1 of the HAX1 gene, results from an A to G substitution at nucleotide position 14. The aspartic acid at codon 5 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
