In summary, this variant is a novel missense change with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C65"). This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a LMNA-related disease. This sequence change replaces isoleucine with serine at codon 297 of the LMNA protein (p.Ile297Ser). The isoleucine residue is highly conserved and there is a large physicochemical difference between isoleucine and serine. -

The p.I297S variant (also known as c.890T>G), located in coding exon 5 of the LMNA gene, results from a T to G substitution at nucleotide position 890. The isoleucine at codon 297 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
