The p.R40W variant (also known as c.118C>T), located in coding exon 3 of the SDHC gene, results from a C to T substitution at nucleotide position 118. The arginine at codon 40 is replaced by tryptophan, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This sequence change replaces arginine, which is basic and polar, with tryptophan, which is neutral and slightly polar, at codon 40 of the SDHC protein (p.Arg40Trp).
