This variant reduces the quality of the splice donor site in intron 4, but is not predicted to appreciably affect splicing by in silico modeling. The c.606+4A>G intronic substitution occurs at a position that is conserved across mammalian species. This variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.606+4A>G as a variant of uncertain significance. -
