This sequence change replaces proline, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 4090 of the USH2A protein (p.Pro4090Thr). This variant is present in population databases (rs780893919, gnomAD 0.002%). This missense change has been observed in individual(s) with clinical features of retinitis pigmentosa or Usher syndrome (PMID: 23591405; internal data). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 550401).
