Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 25999674, 26927203, 27318125; Invitae). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This missense change has been observed in individual(s) with Usher syndrome (PMID: 27460420). This sequence change replaces proline, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 2241 of the USH2A protein (p.Pro2241Thr). This variant is present in population databases (no rsID available, gnomAD 0.0009%). -
