This variant was determined to be likely pathogenic according to ACMG Guidelines, 2015 [PMID:25741868]. This variant has been previously reported as a de novo variant in a patient with possible nemaline myopathy [PMID 19562689] -
This sequence change replaces proline, which is neutral and non-polar, with arginine, which is basic and polar, at codon 334 of the ACTA1 protein (p.Pro334Arg). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of congenital myopathy (PMID: 32154989; Invitae). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 1031829). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt ACTA1 protein function with a negative predictive value of 80%.
