Together these subunits are composed of 4 RNA species and approximately 80 structurally distinct proteins. This gene encodes a ribosomal protein that is a component of the 60S subunit. The protein belongs to the L5P family of ribosomal proteins. It is located in the cytoplasm. The protein probably associates with the 5S rRNA. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. This alteration was first reported as a de novo mutation in a 3 month old female with Diamond-Blackfan anemia; this infant did not have congenital malformations and did not respond to steroid therapy (Gazda HT et al. Am J Hum Genet. 2008;83(6):769-80). In addition to the clinical data presented in the literature, alterations that disrupt the canonical splice donor site are typically deleterious in nature (ACMG Recommendations for Standards for Interpretation and Reporting of Sequence Variations. Revision 2007. Genet Med. 2008;10:294). -
