This sequence change falls in intron 39 of the RYR2 gene. It does not directly change the encoded amino acid sequence of the RYR2 protein. It affects a nucleotide within the consensus splice site. This variant is present in population databases (rs376389213, gnomAD 0.02%). This variant has been observed in individual(s) with early repolarization syndrome (PMID: 36138163). ClinVar contains an entry for this variant (Variation ID: 222789). Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant is not likely to affect RNA splicing. The c.6022+5G>A intronic variant results from a G to A substitution 5 nucleotides after coding exon 39 in the RYR2 gene. This variant was reported in an individual with features consistent with early repolarization syndrome (Rayani K et al. Eur J Hum Genet, 2023 May;31:512-520). This nucleotide position is poorly conserved in available vertebrate species. In silico splice site analysis for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
