The encoded protein is one of the components of a calcium channel, composed of a tetramer of the ryanodine receptor proteins and a tetramer of FK506 binding protein 1B proteins, that supplies calcium to cardiac muscle. Mutations in this gene are associated with stress-induced polymorphic ventricular tachycardia and arrhythmogenic right ventricular dysplasia. [provided by RefSeq, Jul 2008]

The glycine at codon 3225 is replaced by serine, an amino acid with similar properties. This alteration was seen in one individual from a sudden cardiac death cohort, who also carried a missense alteration in MYBPC3 (Bagnall RD et al. N. Engl. J. Med., 2016 Jun;374:2441-52). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
