This sequence change replaces serine with glycine at codon 4054 of the RYR2 protein (p.Ser4054Gly). The serine residue is highly conserved and there is a small physicochemical difference between serine and glycine. This variant is present in population databases (rs376781046, ExAC 0.002%). This missense change has been observed in individual(s) with clinical features of RYR2-related conditions (Invitae). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C55". The glycine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. The serine at codon 4054 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
