This variant is present in population databases (no rsID available, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with FUCA1-related conditions. ClinVar contains an entry for this variant (Variation ID: 1328977). This variant disrupts a region of the FUCA1 protein in which other variant(s) (p.Lys431Glnfs*27) have been determined to be pathogenic (PMID: 36082656). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic. -

Truncations downstream of this position have not been classified as pathogenic by our laboratory but at-least one downstream extention variant, c.1399T>A (p.*467K), has been reported in the HGMD database and another downstream variant, c.1295G>A (p.Trp432*) has been classified as a VUS by a submitting laboratory in the ClinVar database. The variant allele was found at a frequency of 4e-06 in 251364 control chromosomes. To our knowledge, no occurrence of c.1285_1286insT in individuals affected with Fucosidosis and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as VUS-possibly pathogenic. -
