This sequence change replaces leucine with proline at codon 244 of the FH protein (p.Leu244Pro). The leucine residue is highly conserved and there is a moderate physicochemical difference between leucine and proline. - |

The leucine at codon 244 is replaced by proline, an amino acid with similar properties. This alteration has been observed in at least one individual with a personal and/or family history that is consistent with FH-related disease (Ambry internal data). Based on internal structural analysis, this variant is moderately destabilizing to the local structure (Ambry internal data). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
