This sequence change replaces glutamine, which is neutral and polar, with proline, which is neutral and non-polar, at codon 185 of the FH protein (p.Gln185Pro). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with multiple cutaneous leiomyomas (Invitae). ClinVar contains an entry for this variant (Variation ID: 574229). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. This variant disrupts the p.Gln185 amino acid residue in FH. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 11865300, 21445611, 28300276; Invitae).
