This sequence change replaces methionine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 661 of the NLRP3 protein (p.Met661Arg). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with clinical features of cryopyrin-associated periodic syndrome (Invitae). ClinVar contains an entry for this variant (Variation ID: 643124). This variant disrupts the p.Met661 amino acid residue in NLRP3. Other variant(s) that disrupt this residue have been observed in individuals with NLRP3-related conditions (PMID: 16081838, 24773462), which suggests that this may be a clinically significant amino acid residue. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively.
