Variant summary: TRIM63 c.1052-21_1052-20delGT alters a nucleotide located at a position not widely known to affect splicing. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 0.00015 in 119562 control chromosomes (gnomAD). This frequency is not significantly higher than estimated for a pathogenic variant in TRIM63 causing Hypertrophic Cardiomyopathy (0.00015 vs 0.005), allowing no conclusion about variant significance. To our knowledge, no occurrence of c.1052-21_1052-20delGT in individuals affected with Hypertrophic Cardiomyopathy and no experimental evidence demonstrating its impact on protein function have been reported.
