MUTYH (HGNC:7527): (mutY DNA glycosylase) This gene encodes a DNA glycosylase involved in oxidative DNA damage repair. The enzyme excises adenine bases from the DNA backbone at sites where adenine is inappropriately paired with guanine, cytosine, or 8-oxo-7,8-dihydroguanine, a major oxidatively damaged DNA lesion. The protein is localized to the nucleus and mitochondria. This gene product is thought to play a role in signaling apoptosis by the introduction of single-strand breaks following oxidative damage. Mutations in this gene result in heritable predisposition to colorectal cancer, termed MUTYH-associated polyposis (MAP). Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Apr 2017]

The arginine at codon 247 is replaced by glutamine, an amino acid with highly similar properties. This alteration was identified once in a cohort of 405 patients with at least 10 colonic polyps (Guarinos C et al. Clin. Cancer Res., 2014 Mar;20:1158-68), and in a cohort of 31 Japanese patients with suspected hereditary polyposis (Takao M et al. Int J Clin Oncol, 2018 Jun;23:497-503). This alteration was also detected in a cohort of 118 individuals with features suggestive of inherited renal cell carcinoma (Smith PS et al. Genes Chromosomes Cancer, 2021 Jan;60:5-16). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive.
