The alanine at codon 227 is replaced by valine, an amino acid with similar properties. In a study assessing the base excision repair capabilities of several MUTYH variants, this alteration was found to be partially defective (Komine K et al. Hum. Mutat. 2015 Jul;36:704-11). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
