In summary, this variant is a novel missense change with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies. This variant has not been reported in the literature in individuals with MUTYH-related disease. Familial testing has shown that this variant co-occurs in trans with a MUTYH pathogenic missense variant in an individual affected with colon cancer (Invitae). ClinVar contains an entry for this variant (Variation ID: 406865). This variant is not present in population databases (ExAC no frequency). This sequence change replaces glycine with aspartic acid at codon 176 of the MUTYH protein (p.Gly176Asp). The glycine residue is highly conserved and there is a moderate physicochemical difference between glycine and aspartic acid. -

The p.G176D variant (also known as c.527G>A), located in coding exon 7 of the MUTYH gene, results from a G to A substitution at nucleotide position 527. The glycine at codon 176 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear. -
