Variant summary
Our verdict is Likely pathogenic. Variant got 8 ACMG points: 8P and 0B. PM1PM2PP3_Strong
The NM_001048174.2(MUTYH):āc.253T>Cā(p.Trp85Arg) variant causes a missense change. The variant allele was found at a frequency of 0.00000186 in 1,613,986 control chromosomes in the GnomAD database, with no homozygous occurrence. In-silico tool predicts a pathogenic outcome for this variant. Variant has been reported in ClinVar as Uncertain significance (ā ā ). Another nucleotide change resulting in same amino acid change has been previously reported as Uncertain significancein ClinVar. Another variant affecting the same amino acid position, but resulting in a different missense (i.e. W85G) has been classified as Uncertain significance.

Familial adenomatous polyposis 2 Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Jul 28, 2023 | In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces tryptophan, which is neutral and slightly polar, with arginine, which is basic and polar, at codon 113 of the MUTYH protein (p.Trp113Arg). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with colorectal cancer and/or polyps who also carried another pathogenic variant in MUTYH (PMID: 28644590). ClinVar contains an entry for this variant (Variation ID: 533303). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. - |
Hereditary cancer-predisposing syndrome Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Ambry Genetics | Nov 18, 2024 | The p.W113R variant (also known as c.337T>C), located in coding exon 3 of the MUTYH gene, results from a T to C substitution at nucleotide position 337. The tryptophan at codon 113 is replaced by arginine, an amino acid with dissimilar properties. In one study, this alteration was detected in conjunction with MUTYH p.Y179C alteration in two individuals reported to have polyposis (McVeigh TP et al. Ir Med J, 2016 Dec;109:485). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. - |
