The p.R10P variant (also known as c.29G>C), located in coding exon 1 of the MUTYH gene, results from a G to C substitution at nucleotide position 29. The arginine at codon 10 is replaced by proline, an amino acid with dissimilar properties. This variant has been confirmed in trans with a MUTYH pathogenic variant in an individual with colon polyposis (Ambry internal data). This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear. -
