This sequence change replaces asparagine, which is neutral and polar, with lysine, which is basic and polar, at codon 815 of the MYSM1 protein (p.Asn815Lys). This variant is present in population databases (rs772963146, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with MYSM1-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt MYSM1 protein function with a negative predictive value of 80%.
