Inborn genetic diseases Uncertain:1
The c.988A>G (p.I330V) alteration is located in exon 9 (coding exon 9) of the DOCK7 gene. This alteration results from a A to G substitution at nucleotide position 988, causing the isoleucine (I) at amino acid position 330 to be replaced by a valine (V). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
Developmental and epileptic encephalopathy, 23 Uncertain:1
This variant is present in population databases (rs143611920, gnomAD 0.04%). This sequence change replaces isoleucine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 330 of the DOCK7 protein (p.Ile330Val). This variant has not been reported in the literature in individuals affected with DOCK7-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. ClinVar contains an entry for this variant (Variation ID: 475174). -
