Genes affected
PLEKHG5 (HGNC:29105): (pleckstrin homology and RhoGEF domain containing G5) This gene encodes a protein that activates the nuclear factor kappa B (NFKB1) signaling pathway. Mutations in this gene are associated with autosomal recessive distal spinal muscular atrophy. Multiple transcript variants encoding different isoforms have been found for this gene. The c.1535T>A (p.V512D) alteration is located in exon 14 (coding exon 13) of the PLEKHG5 gene. This alteration results from a T to A substitution at nucleotide position 1535, causing the valine (V) at amino acid position 512 to be replaced by an aspartic acid (D). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

This sequence change replaces valine, which is neutral and non-polar, with aspartic acid, which is acidic and polar, at codon 512 of the PLEKHG5 protein (p.Val512Asp). This variant is present in population databases (rs548243654, gnomAD 0.09%). This variant has not been reported in the literature in individuals affected with PLEKHG5-related conditions. ClinVar contains an entry for this variant (Variation ID: 536769). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
