The p.K510E variant (also known as c.1528A>G), located in coding exon 11 of the NEXN gene, results from an A to G substitution at nucleotide position 1528. The lysine at codon 510 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This sequence change replaces lysine, which is basic and polar, with glutamic acid, which is acidic and polar, at codon 510 of the NEXN protein (p.Lys510Glu). This variant is present in population databases (rs727504758, gnomAD 0.002%). This variant has not been reported in the literature in individuals affected with NEXN-related conditions. ClinVar contains an entry for this variant (Variation ID: 179277).
