The proline at codon 611 is replaced by threonine, an amino acid with highly similar properties. This variant has been detected in an individual with dilated cardiomyopathy (DCM) (Hassel D et al. Nat Med, 2009 Nov;15:1281-8). Zebrafish embryos expressing the corresponding variant recapitulated DCM phenotype (Hassel D et al. Nat Med, 2009 Nov;15:1281-8). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear. - |
