Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251376 control chromosomes (gnomAD). c.2983G>T has been reported in the literature in the homozygous state in an individual affected with Dihydropyrimidine Dehydrogenase Deficiency (Vreken_1998). At least two publications report experimental evidence evaluating an impact on protein function and found that the variant results in <12.5% enzymatic activity compared to the WT protein (e.g. Vreken_1998, Offer_2014). The following publications have been ascertained in the context of this evaluation (PMID: 24648345, 9686374).
