ClinVar contains an entry for this variant (Variation ID: 565556). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. The p.A56S variant (also known as c.166G>T), located in coding exon 1 of the SUFU gene, results from a G to T substitution at nucleotide position 166. The alanine at codon 56 is replaced by serine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
