The p.D351H variant (also known as c.1051G>C), located in coding exon 2 of the RBM20 gene, results from a G to C substitution at nucleotide position 1051. The aspartic acid at codon 351 is replaced by histidine, an amino acid with similar properties. This alteration has been reported in a genetic testing cohort with limited clinical details (Parikh VN et al. Circ Heart Fail, 2019 03;12:e005371). This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
