Testing was performed at Inviatae. Given the lack of case data, we consider this variant a variant of uncertain significance and we do not feel it is suitable for assessing risk in healthy relatives ("predictive genetic testing"). There are no reported cases in the literature (per PubMed and Google). In silico analyses suggest this missense change would be tolerated. However, per the lab report, "Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies." The variant is present in 2 of 90936 individuals listed in the Genome Aggregation Consortium Dataset (gnomAD; http://gnomad.broadinstitute.org/), which currently includes variant calls on >140,000 unrelated individuals of African, Asian, European, Latino, and Ashkenazi descent. Specifically the variant has been seen in 2 of 36378 European individuals (MAF = 0.002749%). The site is well covered. - |
