Variant summary
Our verdict is Uncertain significance. The variant received 2 ACMG points: 2P and 0B. PM2
The NM_002313.7(ABLIM1):c.1997G>A(p.Arg666Gln) variant causes a missense, splice region change involving the alteration of a conserved nucleotide. The variant allele was found at a frequency of 0.0000112 in 1,613,028 control chromosomes in the GnomAD database, with no homozygous occurrence. 2/3 splice prediction tools predict no significant impact on normal splicing. Variant has been reported in ClinVar as Uncertain significance (★). Another variant affecting the same amino acid position, but resulting in a different missense (i.e. R666W) has been classified as Uncertain significance.

not specified Uncertain:1
The c.1997G>A (p.R666Q) alteration is located in exon 18 (coding exon 18) of the ABLIM1 gene. This alteration results from a G to A substitution at nucleotide position 1997, causing the arginine (R) at amino acid position 666 to be replaced by a glutamine (Q). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
