The proline at codon 16 is replaced by alanine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. This variant has been described in a single individual affected with sporadic ALS but zygosity of the alteration is unclear (Lattante S et al. Neurology, 2012 Jul;79:66-72). In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
