The encoded protein binds actin and interacts with thin filaments and Z-line associated proteins in striated muscle. This protein may be involved in cardiac myofibril assembly. A shorter isoform of this protein termed LIM nebulette is expressed in non-muscle cells and may function as a component of focal adhesion complexes. The Ala726Thr variant in NEBL has not been reported in the literature nor previo usly identified by our laboratory. Alanine (Ala) at position 726 is not conserve d in mammals (dog has a glycine, Gly), suggesting a change at this position may be tolerated. Additional computational analyses (biochemical amino acid properti es, AlignGVGD, PolyPhen2, and SIFT) do not provide strong support for or against an impact to the protein. Additional information is needed to fully assess the clinical significance of this variant.
