The p.R313W variant (also known as c.937C>T), located in coding exon 5 of the RET gene, results from a C to T substitution at nucleotide position 937. The arginine at codon 313 is replaced by tryptophan, an amino acid with dissimilar properties. This variant has been previously reported in Spanish individuals with Hirschsprung disease (Núñez-Torres et al., BMC Med. Genet. 2011 Oct;12:138; Luzón-Toro B et al. PLoS ONE, 2013 Jan;8:e54800). This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
