This sequence change replaces glutamine, which is neutral and polar, with glutamic acid, which is acidic and polar, at codon 681 of the RET protein (p.Gln681Glu). This variant is present in population databases (rs567241943, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with RET-related conditions. ClinVar contains an entry for this variant (Variation ID: 477337). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.2041C>G has been reported in the literature in individual(s) affected with complex congenital heart disease (example, Chen_2020). These report(s) do not provide unequivocal conclusions about association of the variant with Multiple Endocrine Neoplasia Type 2. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
