This sequence change replaces tryptophan, which is neutral and slightly polar, with arginine, which is basic and polar, at codon 1056 of the RET protein (p.Trp1056Arg). This variant is present in population databases (rs373423271, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with RET-related conditions. ClinVar contains an entry for this variant (Variation ID: 543728). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. The p.W1056R variant (also known as c.3166T>C), located in coding exon 19 of the RET gene, results from a T to C substitution at nucleotide position 3166. The tryptophan at codon 1056 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
