Defects in this gene are associated with Charcot-Marie-Tooth disease type 1D (CMT1D), Charcot-Marie-Tooth disease type 4E (CMT4E), and with Dejerine-Sottas syndrome (DSS). This variant has been reported to affect EGR2 protein function (PMID: 17717711, 27013732). This variant has been observed in individual(s) with autosomal dominant Dejerine-Sottas syndrome (PMID: 17717711). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 16753). This variant is not present in population databases (ExAC no frequency). This sequence change replaces glutamic acid with lysine at codon 412 of the EGR2 protein (p.Glu412Lys). The glutamic acid residue is highly conserved and there is a small physicochemical difference between glutamic acid and lysine. - |
