The c.674G>T (p.C225F) alteration is located in exon 6 (coding exon 5) of the CTNNA3 gene. This alteration results from a G to T substitution at nucleotide position 674, causing the cysteine (C) at amino acid position 225 to be replaced by a phenylalanine (F). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

This sequence change replaces cysteine, which is neutral and slightly polar, with phenylalanine, which is neutral and non-polar, at codon 225 of the CTNNA3 protein (p.Cys225Phe). This variant is present in population databases (rs747835139, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with CTNNA3-related conditions. ClinVar contains an entry for this variant (Variation ID: 572206). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt CTNNA3 protein function with a negative predictive value of 80%.
