Z-lines in striated muscle are a sign of this organization, representing the ends of actin thin filaments, titin, nebulin or nebulette and accessory proteins required for structure and function. This gene encodes a protein which interacts with nebulin in skeletal muscle or nebulette in cardiac muscle and alpha-actinin. In addition, this gene product can interact with a protein with the I-band indicating it has a regulatory as well as structural function. Alternative splicing results in multiple transcript variants. The arginine at codon 810 is replaced by cysteine, an amino acid with highly dissimilar properties. This variant has been detected in a non-compaction cardiomyopathy cohort and in an individual with hypertrophic cardiomyopathy; however, clinical details were limited (van Waning JI et al. J. Am. Coll. Cardiol., 2018 Feb;71:711-722; van Lint FHM et al. Neth Heart J. 2019 Jun;27(6):304-309). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
