Isoleucine (Ile) at position 815 is not conserv ed in mammals or evolutionarily distant species and 2 species (manatee and rock pigeon) carry a valine (Val) at this position, raising the possibility that this change may be tolerated. Additional computational prediction tools suggest that the p.Ile815Val variant may not impact the protein, though this information is not predictive enough to rule out pathogenicity. In summary, while the clinical significance of the p.Ile815Val variant is uncertain, the lack of evolutionary c onservation suggest that it is more likely to be benign. -

This variant is not present in population databases (ExAC no frequency). This sequence change replaces isoleucine with valine at codon 815 of the MYPN protein (p.Ile815Val). The isoleucine residue is weakly conserved and there is a small physicochemical difference between isoleucine and valine. This variant has not been reported in the literature in individuals affected with MYPN-related conditions. ClinVar contains an entry for this variant (Variation ID: 229034).
