Z-lines in striated muscle are a sign of this organization, representing the ends of actin thin filaments, titin, nebulin or nebulette and accessory proteins required for structure and function. This gene encodes a protein which interacts with nebulin in skeletal muscle or nebulette in cardiac muscle and alpha-actinin. In addition, this gene product can interact with a protein with the I-band indicating it has a regulatory as well as structural function. Alternative splicing results in multiple transcript variants. Variant classified as Uncertain Significance - Favor Benign. The p.Ile815Val var iant in MYPN has not been previously reported in individuals with cardiomyopathy or in large population studies. Isoleucine (Ile) at position 815 is not conserv ed in mammals or evolutionarily distant species and 2 species (manatee and rock pigeon) carry a valine (Val) at this position, raising the possibility that this change may be tolerated. Additional computational prediction tools suggest that the p.Ile815Val variant may not impact the protein, though this information is not predictive enough to rule out pathogenicity. In summary, while the clinical significance of the p.Ile815Val variant is uncertain, the lack of evolutionary c onservation suggest that it is more likely to be benign. -

This variant is not present in population databases (ExAC no frequency). This sequence change replaces isoleucine with valine at codon 815 of the MYPN protein (p.Ile815Val). The isoleucine residue is weakly conserved and there is a small physicochemical difference between isoleucine and valine. This variant has not been reported in the literature in individuals affected with MYPN-related conditions. ClinVar contains an entry for this variant (Variation ID: 229034). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
